Evaluation of Seroquel (quetiapine) and Seroquel XR (quetiapine fumarate) in the treatment of schizophrenia and bipolar disorder: a double-blind, placebo-controlled study. Case reports. New York, NY: Eli Lilly and Company; 2002.
The results of a double-blind, placebo-controlled clinical trial (C-DCT) comparing the efficacy of Seroquel (quetiapine) and Seroquel XR (quetiapine fumarate) in the treatment of schizophrenia and bipolar disorder are presented. Participants were adults with a diagnosis of schizophrenia. They were treated with either Seroquel XR (quetiapine fumarate) or a placebo for 7 to 10 weeks with or without an additional therapy for a total of 10 to 12 weeks. The primary outcome measure was change from baseline in the Global Severity of Illness scale, and the secondary outcome measures were change from baseline in the Clinical Global Impression Scale, the change from baseline in the Global Assessment Scale, and the change from baseline in the Clinical Global Impression. Safety data were obtained from at least 5 of the 6 randomized, double-blind, placebo-controlled, open-label trials. Data were analyzed using Monte Carlo simulations (NCT00346617). Data were analyzed with a two-sided Student’s t-test and one-way analysis of variance (ANOVA) with post hoc analysis. The study group received placebo for 7 to 10 weeks. In addition, the dose of Seroquel XR was lowered by a dose-related increase in serum levels of both drugs. The dose-response relationship was assessed by the change from baseline in the Global Assessment Scale and the change from baseline in the Global Assessment Scale. Safety data from the 6 randomized, double-blind, placebo-controlled open-label trials were analyzed with a one-way ANOVA with post hoc analysis. There was no difference in the safety data between the Seroquel XR group and the placebo group. Data were analyzed with a one-way ANOVA with post hoc analysis. Safety data were analyzed with a one-way ANOVA with post hoc analysis. Safety data were analyzed with a two-sided Student’s t-test.
The primary efficacy measure of Seroquel (quetiapine fumarate) and Seroquel XR (quetiapine fumarate) was the Positive and Negative Syndrome Scale (PANSS). The PANSS scale used to measure depression was administered at the start of the study and at the end of the study. A score of 0 (never) or 2 (daily) was assigned if there was a 1 or 2 on the PANSS scale score. A score of 3 (daily) or 4 (every 4 to 8 hours) was assigned if there was a 3 or 4 on the PANSS scale score. A score of 5 or 6 was assigned if there was a 5 or 6 on the PANSS scale score. A score of 7 or 8 was assigned if there was a 7 or 8 on the PANSS scale score. A score of 9 or higher was assigned if there was a 9 or higher PANSS scale score.
